Upright microscopes are used for phase contrast or wide field fluorescence microscopy of samples or living cells that are pressed within a cover slip and a slide. The microscopy of tissue sections or fixed cells is the additional application. For live-cell imaging, particularly with dipping objective lenses, these microscopes can be used. This kind of method is normally practiced for larger samples like animals and tissue. The working distance in an upright microscope is more, which leads to lower resolution and weaker fluorescence signals. Upright microscopes are not suitable for the use of objectives with high numerical aperture directly on the cover slip bottom, on which the cells adhere.
